Description
Blood PCR Enhancer (2X), also known as Blood-Resistant PCR Enhancer (2X), is a PCR enhancer used with DirectTaq™ HF DNA Polymerase (Blood-Resistant) to overcome PCR amplification obstacles caused by high GC content in templates. Amplifying high GC content DNA fragments has always been a challenge in molecular biology operations. This product can be used for direct PCR reactions containing anticoagulants such as heparin, sodium citrate, and EDTA, significantly improving the amplification efficiency and specificity of high GC content DNA templates. This product can also be used for amplifying high GC content target fragments in regular PCR.
Blood PCR Enhancer (2X) is compatible with various commercially available blood-resistant DNA polymerases, such as our DirectTaq™ HF DNA Polymerase (Blood-Resistant) and NEB’s Hemo KlenTaq.

Precautions
- When using this product, Blood PCR Enhancer (2X), you must also use a PCR Buffer at a final concentration of 1X that is compatible with the blood-resistant DNA polymerase.
- If there are crystalline precipitates when dissolving this product, incubate briefly in a 50°C water bath to promote dissolution, and ensure it is completely dissolved before use.
- Please use freshly collected anticoagulated blood samples for testing; if using frozen samples, avoid repeated freeze-thaw cycles to prevent fragmentation and degradation of the target gene during the process.
- When setting up the PCR reaction system, the recommended amount of anticoagulated blood is about 5-10% of the total PCR system volume. It can be added directly to the PCR system without any additional treatment.
- After the PCR reaction, it is recommended to centrifuge at 3000-5000g for 3-5 minutes to precipitate blood cell debris, making it easier to collect the supernatant for electrophoresis analysis, etc. Note: Due to the nature of anticoagulated blood, a transparent gel-like substance may appear at the bottom of the PCR tube after the PCR reaction, which is normal.
- Since PCR reactions are highly sensitive and can amplify the target gene sequence over 10 million times, please take care to avoid contamination with trace amounts of DNA to be amplified. It is advisable to set up a blank control without a template to confirm whether there is contamination of the DNA to be amplified.
