Description
RNA pull-down is a primary method for detecting interactions between RNA and RNA binding proteins (RBPs). The biotin-RNA pull-down kit uses biotin-labeled RNA transcribed in vitro to mimic natural RNA molecules as probes, which incubate with sample lysates. The probes form RNA-protein complexes with proteins in the lysate. The biotin-labeled RNA is specifically captured through the strong affinity between streptavidin magnetic beads and biotin, allowing for the enrichment of RBPs. Specific proteins can then be detected using Western Blot, or unknown proteins can be identified using mass spectrometry (LC-MS/MS).
Applications
RNA binding proteins (RBPs) are powerful and widely-ranging regulatory factors, comprising about 5%-10% of all cellular proteins. While a few RNAs function independently as ribozymes, most RNA performs its roles through interactions with proteins to form RNA-protein complexes. On one hand, RBPs are involved in RNA regulation, while on the other, RNA regulates RBPs. The F2-RNA pull-down kit can be applied in the following research areas:
- RNA activity, transport, and localization regulation;
- RNA stability, translation, and degradation;
- RNA synthesis and alternative splicing;
- RNA modifications;
- RBP activity, localization, or interactions with other proteins.

Brief Usage Procedure
- Amplify the target sequence with a T7 promoter and use it as a template to prepare biotin-labeled RNA probes.

- Incubate with the protein sample to retrieve proteins binding to the target RNA.
