Description
In normal cells, phosphatidylserine (PS) is distributed only on the inner side of the lipid bilayer of the cell membrane. However, in the early stages of apoptosis, phosphatidylserine (PS) flips from the inner side to the outer side of the lipid bilayer. Annexin V is a 35-36 kDa Ca²⁺-dependent phospholipid-binding protein with a high affinity for phosphatidylserine. Therefore, it can bind to the cell membrane of early apoptotic cells through the exposed phosphatidylserine on the outer side of the cell. Consequently, Annexin V is used as a sensitive marker for detecting early apoptosis. When Annexin V is labeled with fluorescein FITC, the labeled Annexin V serves as a fluorescent probe. The occurrence of apoptosis can be detected using a fluorescence microscope or flow cytometer.
Propidium iodide (PI) is a nucleic acid dye that cannot pass through intact cell membranes. However, in late apoptotic and dead cells, PI can penetrate the cell membrane and stain the nucleus red. Therefore, using Annexin V in conjunction with PI allows for the differentiation of cells at different stages of apoptosis.
This kit is suitable for detecting apoptosis in cultured cells (not recommended for detecting tissue samples).

Additional Notes
- Please centrifuge the micro-reagents to collect the liquid before use.
- Store and use Annexin V-FITC and Propidium Iodide (PI) protected from light.
- Propidium Iodide (PI) is toxic; please wear gloves when handling it.
- This kit is suitable for detecting live cells. When using a flow cytometer, the cell count should not be lower than 1x105. It is not recommended for detecting tissue samples.
- Suspension culture cells are recommended. If using adherent cells, digest with trypsin without EDTA, as improper digestion may cause false positives. Using a cell scraper can cause cell clumping, which affects detection. Cells digested with trypsin can be stored in PBS containing 2% BSA to prevent further damage.
- Fixation of cells may cause fluorescence quenching; please do not fix the samples.
- Due to the variability in cell types, apoptosis inducers, and detection instruments, the fluorescence compensation for flow cytometry will differ. Therefore, it is recommended to use cells treated with apoptosis inducers as controls for fluorescence compensation adjustment in each experiment.
Storage
Store at 2-8°C for 12 months, protected from light.
