Description
In normal cells, phosphatidylserine (PS) is located exclusively on the inner leaflet of the cell membrane lipid bilayer. However, during the early stages of apoptosis, PS translocates from the inner to the outer leaflet of the cell membrane. Annexin V is a calcium-dependent phospholipid-binding protein with a molecular weight of 35-36 kD that has a high affinity for phosphatidylserine. Consequently, it can bind to the exposed phosphatidylserine on the outer leaflet of the membrane in early apoptotic cells. Therefore, Annexin V is used as a sensitive marker for detecting early apoptosis. Annexin V is labeled with the fluorescent dye APC, and the labeled Annexin V serves as a fluorescent probe to detect apoptosis using fluorescence microscopy or flow cytometry.
SYTOX Green dead cell nucleic acid dye (Ex/Em: 502/525 nm, bound to DNA) is a green nucleic acid dye that easily penetrates damaged cell membranes but cannot pass through the membranes of live cells. Once bound to nucleic acids, its fluorescence intensity increases by more than 500 times. SYTOX Green dead cell nucleic acid dye can be used to stain RNA and DNA in dead eukaryotic cells and is also applicable to Gram-negative bacteria. The dye can be excited with a 488 nm laser (or any other excitation source with a 450-490 nm excitation range) and can be used in combination with Annexin V-APC.
This kit is suitable for detecting apoptosis in cultured cells and is not recommended for use with tissue samples.

Additional Notes
Annexin V-APC components should be aliquoted as needed and stored at -20°C. Keep protected from light during storage and use, and avoid repeated freeze-thaw cycles.
Binding Buffer components can be stored at 4°C when not in use, and should be kept protected from light.

Prior to use, centrifuge micro-reagents to collect liquid.
This kit is suitable for detecting live cells. When using a flow cytometer, the cell count should not be less than 1x105. This kit is not recommended for tissue sample detection.
It is recommended to use suspended cells. For adherent cells, use trypsin without EDTA for digestion. Improper digestion may lead to false positives, while using a cell scraper can cause cell clumping, which may affect detection. Cells can be stored in PBS containing 2% BSA after trypsin digestion to prevent further damage.
Fixation of cells may lead to fluorescence quenching, so please do not fix the samples.
Due to variations in cell type, apoptosis inducers, and detection instruments, fluorescence compensation for flow cytometry may differ. Therefore, it is recommended to use apoptosis-induced cells as controls for each detection to adjust fluorescence compensation.
Storage
Store at 2-8°C for 12 months, protected from light.
