Description
Albumin Assay Kit with BCG, also known as the Albumin Assay Kit with Bromocresol Green or BCG Albumin Assay Kit, is designed for rapid detection of albumin concentration in serum and plasma samples from humans, mice, rats, cattle, and more. This kit uses a colorimetric method based on the binding of albumin with the BCG dye.
Albumin is one of the most abundant and important proteins in human blood. It is synthesized by polyribosomes associated with the endoplasmic reticulum of liver cells but is not stored in the liver. Under normal physiological conditions, only 20-30% of liver cells produce albumin, but its synthesis rate can increase 2-3 times under specific physiological needs. In healthy humans, albumin accounts for about 50% of plasma proteins. It has a molecular weight of 66 kDa, making it smaller and very stable compared to other plasma proteins. Albumin is highly water-soluble, has an oval shape, and features low viscosity properties.
One of its key roles is maintaining plasma colloidal osmotic pressure, with a normal reference range of 3.5-5g/dl (35-50mg/ml). Research has also shown that albumin plays a crucial role in maintaining vascular endothelial homeostasis and provides protection against glycocalyx inflammation and damage. Furthermore, albumin can bind various compounds, aiding in the transport, delivery, and clearance of drugs, as well as the absorption, storage, and disposal of potentially harmful biomolecules. Albumin also helps regulate acid-base balance, inflammation, endothelial integrity, and binding of endogenous and exogenous compounds.

Principle
Under acidic conditions, albumin carries a positive charge and binds to the negatively charged Bromocresol Green (BCG), forming a complex that changes color from yellow to blue-green. This complex has a maximum absorption peak at 628nm. The absorbance at this wavelength is proportional to the albumin concentration, allowing for accurate quantification of albumin in samples. This kit exhibits high specificity and does not react with other plasma proteins such as IgG.
Features
- This kit features a wide detection range (0.04-10g/dl) and is user-friendly. The concentration of albumin in different samples can vary widely. For example, healthy humans typically have albumin concentrations ranging from 3.5-5g/dl. However, patients with hypoalbuminemia or hyperalbuminemia may have albumin concentrations below 3.5g/dl or above 5g/dl. The kit's broad detection range allows for direct testing of most samples without the need for dilution, making it simple and convenient to use. Bovine Serum Albumin (BSA) is provided as a standard to set up the standard curve.
- This kit offers high sensitivity, with a detection limit as low as 0.04g/dl, and a minimum detectable albumin amount of just 2µg.
- This kit offers rapid detection. Only 1-10 minutes of incubation at room temperature is needed to measure absorbance. Typically, 10-20 samples can be tested in under 10 minutes.
Storage
- Store at 4ºC for up to one year.
- BCG Reagent A and BCG Reagent B should be stored protected from light.
- The entire kit can be stored at -20ºC for a longer shelf life.
Precautions
- Fully dissolve the protein standard, mix well, and then dilute to prepare a series of BSA standard solutions of different concentrations.
- Allow all reagents to equilibrate to room temperature before use. After use, store the reagents under the conditions specified in the kit.
- A microplate reader capable of detecting wavelengths between 610-630nm is required, with an optimal detection wavelength of 628nm, along with a 96-well plate. If a microplate reader is unavailable, a standard spectrophotometer can be used. Adjust the volumes of BCG Reagent A and BCG Reagent B as needed to meet the minimum detection volume of the cuvette. The sample and standard volumes can be proportionally increased or remain unchanged. Using a spectrophotometer may significantly reduce the number of samples that can be tested per kit.
