Description
AaCas12b, derived from Alicyclobacillus acidiphilus, is a dual-RNA-guided DNA endonuclease belonging to CRISPR type V-B. It features a shorter PAM sequence (-TTN) compared to Cas12a and exhibits higher spacer specificity (20 nt). Guided by single-stranded RNA, AaCas12b binds to target DNA and, after cleaving the target sequence, continues to efficiently cut other single-stranded nucleic acids within the system. This "trans" activity, which is approximately 10 times higher than its activity on target nucleic acids, can be harnessed for signal amplification in nucleic acid detection using labeled ssDNA reporter probes.
In terms of substrate recognition, AaCas12b targets both double-stranded and single-stranded DNA. Except for the dependence on PAM upstream of the NTS for double-stranded DNA recognition, there is no significant difference in reaction efficiency between double-stranded and single-stranded targets. Compared to Cas12a, AaCas12b has a broader temperature tolerance range, with an optimal reaction temperature of 30-60°C, making it more suitable for simultaneous use with isothermal amplification reactions like LAMP.
AaCas12b can utilize engineered single-molecule guide RNA (sgRNA) for reactions, and through our extensive optimization, the total length of sgRNA can be controlled within 105 nt, making it ideal for high-throughput synthesis. Additionally, AaCas12b can employ single-base mutated tgRNA for highly specific SNP detection.
