Biochemical/Physiological Actions
2-Deoxy-D-glucose (2-DG) is a glucose analog that inhibits glycolysis by acting on hexokinase (the rate-limiting step of glycolysis). It is phosphorylated by hexokinase to 2-DG-P, which cannot be further metabolized by phosphoglucose isomerase. This leads to the accumulation of 2-DG-P in cells and depletion of cellular ATP. In vitro, 2-DG induces autophagy, increases ROS production, and activates AMPK.
