Recombinant Lysyl Endopeptidase, GMP Grade
Description
Lysyl endopeptidase (Lys-C; EC 2.7.6.716.4) was first isolated by Masaki et al. from the soil bacterium Achromobacter lyticus. It specifically recognizes and cleaves peptide bonds at the carboxyl-terminal of lysine residues. This product is sourced from A. lyticus, recombinantly expressed in Escherichia coli, and is tag-free with a molecular weight of 31kDa. Its optimal enzymatic reaction occurs at pH 9.0–10.0 and a temperature range of 30–37°C, with activity decreasing above 50°C. The enzyme activity is stable after incubation at 30°C for 6 hours in 4M urea or 0.2% SDS solutions. Activity is inhibited by DFP, PMSF, and TLCK.
Applications
- Biopharmaceuticals: Enzymatic cleavage or transpeptidation in the production of recombinant insulin and its analogs (e.g., aspart insulin, detemir, and degludec insulin), as well as recombinant peptide drug manufacturing.
- Biological Research: Quality peptide mapping for monoclonal antibodies or biomacromolecular drugs, protein sequencing, and proteomics analysis.
Quality Parameters
- Appearance: White or off-white lyophilized powder
- Purity (SDS-PAGE): ≥95%
- Relative Molecular Weight: 31kDa ± 3.1kDa
- Specific Activity: ≥3.0 AU/mg
- Endotoxin: <1 EU/mg
Notes:
① Additional tests such as microbial limits, host protein residues, or host DNA residues can be included in the CoA upon request.
② Enzyme activity definition: One activity unit (AU) is defined as the amount of enzyme required to catalyze the generation of 1μmol of PNA per minute at 37±2°C in a 0.2M Tris-HCl buffer at pH 9.5.
Advantages
- Produced under GMP-compliant conditions, with batch production scales exceeding 50g and an annual output in kilograms. Production quality is stable across batches.
- High purity, high enzymatic activity, and excellent cleavage specificity with no residual contaminant enzymes. No animal-derived materials are used during production.
Storage
The lyophilized powder remains stable for 24 months when stored below -15°C. After reconstitution, it can be stored below -15°C and endure up to three freeze-thaw cycles. Reconstituted solutions can be stored at 2–8°C for up to 7 days.
