Description
Oligo dT (oligodeoxythymidine) is an oligonucleotide sequence composed of multiple deoxythymidine nucleotides, which specifically binds to the poly(A) tail of eukaryotic mRNA. This makes Oligo dT an essential tool for mRNA detection and purification. Oligo dT has a broad range of applications, including use as a reverse transcription primer and as a key component in mRNA purification affinity materials and Oligo dT magnetic bead preparation.
Oligo dT magnetic beads are magnetic polymer microspheres with carboxyl groups on their surface, conjugated with aminated oligonucleotide single strands. These beads can quickly and efficiently isolate and purify mRNA from total RNA, cell lysates, and in vitro transcribed RNA samples by complementary pairing with the poly(A) tail of eukaryotic mRNA. For mRNA purification, Oligo dT is bonded to the surface of a matrix, allowing the poly(A) tail of mRNA to bind to Oligo dT through hydrogen bonding. This process facilitates impurity removal and large-scale mRNA purification.
Purified mRNA from magnetic beads or affinity materials has numerous applications, including RT-PCR, solid-phase cDNA library construction, next-generation sequencing, primer extension, gene expression analysis, RACE, RNA sequencing, and affinity purification.
