Description
Human UBDUB CRISPR Knockout Library is designed for the knockout and screening of genes involved in ubiquitination and deubiquitination complexes in humans, targeting a total of 1,500 genes. The library comprises 9,274 knockout vectors, with each gene targeted by six gRNA knockout vectors. Additionally, there are 1,000 control vectors (including 1,000 targeting non-genic sequences). This library utilizes the YKO-LV004 backbone, a dual-plasmid vector system that only expresses gRNA, with the Cas9 gene on a separate vector, requiring paired usage.

Oligonucleotides with BsmB1 restriction sites for guide RNAs were synthesized by SBS Genetech Co., Ltd., then phosphorylated using T4 kinase (31). The phosphorylated oligonucleotides were cloned into LentiCRISPR v2 (Plasmid no. 52961; Addgene) and the sequences of the cloned plasmids that were extracted from numerous selected colonies were confirmed by SBS Genetech Co., Ltd.
