Human CRISPR Whole Genome Knockout Library (Single Vector System A+B)
Description
This knockout library is suitable for whole-genome knockout and screening in humans, targeting 19,050 human genes and comprising a total of 123,411 knockout vectors. Each gene is targeted by six gRNA knockout vectors, with an additional 1,000 control vectors targeting non-genomic sequences. The library uses the lentiCRISPR v2 backbone and is an all-in-one vector system, meaning the gRNA and Cas9 gene are on the same vector and can be used independently.

Oligonucleotides with BsmB1 restriction sites for guide RNAs were synthesized by SBS Genetech Co., Ltd., then phosphorylated using T4 kinase (31). The phosphorylated oligonucleotides were cloned into LentiCRISPR v2 (Plasmid no. 52961; Addgene) and the sequences of the cloned plasmids that were extracted from numerous selected colonies were confirmed by SBS Genetech Co., Ltd.
