Human CRISPR Whole Genome Activation Library
Description
This activation library is suitable for whole-genome activation and screening in humans, targeting 23,430 human genes and comprising a total of 70,290 knockout vectors. Each gene is targeted by three gRNA knockout vectors. The library uses the lenti sgRNA(MS2)_puro optimized backbone and is an all-in-one vector system, meaning the gRNA and Cas9 gene are on the same vector and can be used independently.

Oligonucleotides with BsmB1 restriction sites for guide RNAs were synthesized by SBS Genetech Co., Ltd., then phosphorylated using T4 kinase (31). The phosphorylated oligonucleotides were cloned into LentiCRISPR v2 (Plasmid no. 52961; Addgene) and the sequences of the cloned plasmids that were extracted from numerous selected colonies were confirmed by SBS Genetech Co., Ltd.
